Evaluation of critical weed-free period for three sweetpotato (Ipomoea batatas) cultivars

Sweetpotato [Ipomoea batatas (L.) Lam.] is a staple crop that provides nutritional benefits to humans globally, but it is subject to yield loss when competing with weeds, especially during the early stage of establishment. Yield loss can vary widely based on the cultivar, production environment, weed species, and management techniques. To address this challenge, we conducted field research at the Samuel G. Meigs Horticulture Research Farm, Lafayette, IN, and at the Southwest Purdue Agricultural Center, Vincennes, IN, in 2022 to determine the effect of sweetpotato cultivar on the critical weed-free period. The experiment was a split-plot design, with weed-free interval treatments as the main plot factor and cultivar as the subplot factor. The three cultivars used were ‘Covington’, ‘Monaco’, and ‘Murasaki’. Weeds were removed by hand and allowed to establish and compete with the crop beginning at 0, 14, 21, 28, 35, or 42 d after transplanting (DAP). As the weed-free interval increased from 0 to 42 DAP, predicted total yield increased from 19 kg ha−1 to 20,540 kg ha−1 for Covington, 3 kg ha−1 to 11,407 kg ha−1 for Monaco, and 125 kg ha−1 to 13,460 kg ha−1 for Murasaki at the Lafayette location. At Vincennes, as the weed-free interval increased from 0 to 42 DAP, predicted total yield increased from 14,664 kg ha−1 to 33,905 kg ha−1 for Covington, 4,817 kg ha−1 to 18,059 kg ha−1 for Monaco, and 12,735 kg ha−1 to 21,105 kg ha−1 for Murasaki. A threshold of ≤10% total yield reduction was achieved by maintaining sweetpotatoes weed-free 24 DAP for Covington, 20 DAP for Murasaki, and 33 DAP for Monaco.
Design and implementation of a digital site-less clinical study of serial rapid antigen testing to identify asymptomatic SARS-CoV-2 infection

Background:
Rapid antigen detection tests (Ag-RDT) for SARS-CoV-2 with emergency use authorization generally include a condition of authorization to evaluate the test’s performance in asymptomatic individuals when used serially. We aim to describe a novel study design that was used to generate regulatory-quality data to evaluate the serial use of Ag-RDT in detecting SARS-CoV-2 virus among asymptomatic individuals.
Methods:This prospective cohort study used a siteless, digital approach to assess longitudinal performance of Ag-RDT. Individuals over 2 years old from across the USA with no reported COVID-19 symptoms in the 14 days prior to study enrollment were eligible to enroll in this study. Participants throughout the mainland USA were enrolled through a digital platform between October 18, 2021 and February 15, 2022. Participants were asked to test using Ag-RDT and molecular comparators every 48 hours for 15 days. Enrollment demographics, geographic distribution, and SARS-CoV-2 infection rates are reported.
Key Results:A total of 7361 participants enrolled in the study, and 492 participants tested positive for SARS-CoV-2, including 154 who were asymptomatic and tested negative to start the study. This exceeded the initial enrollment goals of 60 positive participants. We enrolled participants from 44 US states, and geographic distribution of participants shifted in accordance with the changing COVID-19 prevalence nationwide.
Conclusions:The digital site-less approach employed in the “Test Us At Home” study enabled rapid, efficient, and rigorous evaluation of rapid diagnostics for COVID-19 and can be adapted across research disciplines to optimize study enrollment and accessibility.
GASKAP-HI Pilot Survey Science III: An unbiased view of cold gas in the Small Magellanic Cloud

We present the first unbiased survey of neutral hydrogen absorption in the Small Magellanic Cloud. The survey utilises pilot neutral hydrogen observations with the Australian Square Kilometre Array Pathfinder telescope as part of the Galactic Australian Square Kilometre Array Pathfinder neutral hydrogen project whose dataset has been processed with the Galactic Australian Square Kilometre Array Pathfinder-HI absorption pipeline, also described here. This dataset provides absorption spectra towards 229 continuum sources, a 275% increase in the number of continuum sources previously published in the Small Magellanic Cloud region, as well as an improvement in the quality of absorption spectra over previous surveys of the Small Magellanic Cloud. Our unbiased view, combined with the closely matched beam size between emission and absorption, reveals a lower cold gas faction (11%) than the 2019 ATCA survey of the Small Magellanic Cloud and is more representative of the Small Magellanic Cloud as a whole. We also find that the optical depth varies greatly between the Small Magellanic Cloud’s bar and wing regions. In the bar we find that the optical depth is generally low (correction factor to the optically thin column density assumption of $\mathcal{R}_{\mathrm{HI}} \sim 1.04$ ) but increases linearly with column density. In the wing however, there is a wide scatter in optical depth despite a tighter range of column densities.

Mental, neurological and substance use (MNS) disorders are a leading, but neglected, cause of morbidity and mortality in sub-Saharan Africa. The treatment gap for MNS is vast with only 10% of people with MNS disorders in low-income countries accessing evidence-based treatments. Reasons for this include low awareness of the burden of MNS disorders and limited evidence to support development, adaptation and implementation of effective and feasible treatments. The overall goal of the African Mental Health Research Initiative (AMARI) is to build an African-led network of MNS researchers in Ethiopia, Malawi, South Africa and Zimbabwe, who are equipped to lead high quality mental health research programs that meet the needs of their countries, and to establish a sustainable career pipeline for these researchers with an emphasis on integrating MNS research into existing programs such as HIV/AIDS. This paper describes the process leading to the development of AMARI's objectives through a theory of change workshop, successes and challenges that have been faced by the consortium in the last 4 years, and the future role that AMARI could play in further building MNS research capacity by brining on board more institutions from low- and middle-income countries with an emphasis on developing an evidence-based training curriculum and a research-driven care service.
Feasibility of conducting an early pregnancy diet and lifestyle e-health intervention: the Pregnancy Lifestyle Activity Nutrition (PLAN) project

Background:
Childhood obesity is a global issue. Excessive weight gain in early pregnancy is independently associated with obesity in the next generation. Given the uptake of e-health, our primary aim was to pilot the feasibility of an e-health intervention, starting in the first trimester, to promote healthy lifestyle and prevent excess weight gain in early pregnancy. Methods: Women were recruited between 8 and 11 weeks gestation and randomised to the intervention or routine antenatal care. The intervention involved an e-health program providing diet, physical activity and well-being advice over 12 weeks.
Results:Women (n = 57, 43.9% overweight/obese) were recruited at 9.38 ± 1.12 (control) and 9.06 ± 1.29 (intervention) weeks’ gestation, mainly from obstetric private practices (81.2%). Retention was 73.7% for the 12-week intervention, 64.9% at birth and 55.8% at 3 months after birth.
No difference in gestational weight gain or birth size was detected. Overall treatment effect showed a mean increase in score ranking the perceived confidence of dietary change (1.2 ± 0.46, p = 0.009) and score ranking readiness to exercise (1.21 ± 0.51, p = 0.016) over the intervention. At 3 months, infants weighed less in the intervention group (5405 versus 6193 g, p = 0.008) and had a lower ponderal index (25.5 ± 3.0 versus 28.8 ± 4.0 kg/m3) compared with the control group.
Conclusion and Discussion:A lifestyle intervention starting in the first-trimester pregnancy utilising e-health mode of delivery is feasible. Future studies need strategies to target recruitment of participants of lower socio-economic status and ensure maximal blinding. Larger trials (using technology and focused on early pregnancy) are needed to confirm if decreased infant adiposity is maintained.

In 2006, the Midwest Invasive Plant Network's Research Committee conducted a web-based survey to help identify research needs and interactions between land managers and researchers working to manage invasive plants in the Midwest. Of 192 responses, 30% identified themselves as researchers and 70% identified themselves as managers. Researchers and managers rated working together on invasive plant issues as high or medium in importance, but neither group rated the current level of cooperation as high, with over 90% describing current cooperation as low or medium. Both groups self-associate, with 89% of researchers working with other researchers and 77% of managers working with other managers. “Lack of time” and “lack of money” were the main issues limiting researchers and land managers from working more closely together: money was a greater constraint for researchers and time was more important for land managers. To help researchers and land managers work more effectively together, both groups favored opportunities to develop research-based projects at land managers' sites, with funding from a cooperative grant program. Open-ended responses suggest that on-site experiments and demonstrations of management methods could help researchers and land managers interact more effectively. Researchers rated basic biology as more important than land managers did, but neither group judged testing theories of invasion as a high priority. “Social/political factors” and “risk assessment” were viewed as less important despite their clear relevance in the introduction and spread of invasive plants.
Prevalence of and Risk Factors for Multidrug-Resistant Acinetobacter baumannii Colonization Among High-Risk Nursing Home Residents

OBJECTIVE
To characterize the epidemiology of multidrug-resistant (MDR) Acinetobacter baumannii colonization in high-risk nursing home (NH) residents.
DESIGNNested case-control study within a multicenter prospective intervention trial.
SETTINGFour NHs in Southeast Michigan.
PARTICIPANTSCase patients and control subjects were NH residents with an indwelling device (urinary catheter and/or feeding tube) selected from the control arm of the Targeted Infection Prevention study. Cases were residents colonized with MDR (resistant to ≥3 classes of antibiotics) A. baumannii; controls were never colonized with MDR A. baumannii.
METHODSFor active surveillance cultures, specimens from the nares, oropharynx, groin, perianal area, wounds, and device insertion site(s) were collected upon study enrollment, day 14, and monthly thereafter. A. baumannii strains and their susceptibilities were identified using standard microbiologic methods.
RESULTSOf 168 NH residents, 25 (15%) were colonized with MDR A. baumannii. Compared with the 143 controls, cases were more functionally disabled (Physical Self-Maintenance Score >24; odds ratio, 5.1 [95% CI, 1.8–14.9]; P<.004), colonized with Proteus mirabilis (5.8 [1.9–17.9]; P<.003), and diabetic (3.4 [1.2–9.9]; P<.03). Most cases (22 [88%]) were colonized with multiple antibiotic-resistant organisms and 16 (64%) exhibited co-colonization with at least one other resistant gram-negative bacteria.
CONCLUSIONFunctional disability, P. mirabilis colonization, and diabetes mellitus are important risk factors for colonization with MDR A. baumannii in high-risk NH residents. A. baumannii exhibits widespread antibiotic resistance and a preference to colonize with other antibiotic-resistant organisms, meriting enhanced attention and improved infection control practices in these residents.
